The can is full of refrigerant, in a liquid state and under pressure. When you open the valve, the refrigerant will start flowing out as a gas and push the air in the line out with it. As it keeps flowing out, you connect it to the low pressure line. If you stop the flow of refrigerant before you make the connection, the air will re-enter the line.
It's as good as it can be done with a DIY can.

I don't know how to explain it any better.
