Changing Spark Plugs on RX330 and RX350

I have some questions on changing spark plugs on RX330 and RX350 with drive by wire. By the look, I will have to disconnect the throttle body from the rear intake manifold to reach the rear spark plugs.

I read in a Haynes manual on a 2003 Accord and up, it says that if one removes the throttle body, the drive by wire system needs to be reprogrammed again. I know this is not the same car, but I am concern about this as I don't have a manual.

From the way I see it, one must remove the rear intake manifold to remove the plugs comfortably. I don't see any negative effects in disconnecting the intake manifold and the throttle body by removing the four bolts that links the two parts together.

Can someone shed some light here? Thanks.

The last tune-up on something similar was a 05 3.3 Camry. The upper intake comes off (plastic). I didn't unbolt the TB. I just took everything loose to pick up the upper intake and flip it up towards the battery. I used a bungee to hold it there while I did the rear plugs. There are 3 metal brackets that connect the upper intake to the rear cylinder head. hope that helps.

I would imagine the cowl and wipers may need to be pulled, or at least how it seems like it on a Sienna.
