Engine warm and running.
Remove the seal from the oil sample container, but don’t open it.
Turn the engine off.
Go right under the car and remove the oil drain plug.
Open the sample container and 3/4 fill it from the oil drain stream.

All are listed chronologically, so the one labeled Amsoil (meaning contains Amsoil Engine Flush) is the post-flush UOA. The pre-flush is the UOA to the right of it.

UOA #1 - used oil sampled just prior to adding flush
Flush added and engine run for 20 minutes, then
UOA #2 - taken ~25 minutes after sample 1, contains used oil, Amsoil Engine Flush and whatever the flush was able to clean from internal engine surfaces and suspend in the oil.

And @Escape08 - as @ZeeOSix has clarified, even the cartridge filter ends you referenced are sealed.

I’m guessing the Toyota filter pictured is sealed top and bottom when inserted in the cartridge? The Mann filter ends are clearly sealed.
ST filter ends in the original post are supposed to be sealed. 1/3 of one end is not. That is no buenos.
