NO. A flush in my vocabulary is a VERY short term process which means:

1. Add cleaner to dirty oil and case.
2. bring oil and added cleaner up to operating temperature,
3. drain oil.

For a differential, I would think approx. = 10 miles at high speed should just about do it.
 
It will not hurt to run 500 miles on the neutra. It is not a harsh solvent that would break down the oil, infact as mola has pointed out, it is an ester itself and the amount you're going to use will be very nominal and will have no ill effect on the current base oil. As mola suggested, after running for what ever you want on it, make sure it is hot when you go to drain it.
